叠港公路位于海门区，采用一级公路标准，设计速度为每小时100公里，路基宽度26米，城镇段路基适当加宽。

新建桥涵设计汽车荷载等级采用公路-I级。叠港公路起于海门叠石桥，接星海线，向南经海门工业园、天补、三和后到达并穿越江心沙农场，止于海太汽渡。路线全长约15公里。
